I have a Prusa i3 DIY Kit and the power supply dose match the instructions, so I sent a request from the supplier, and have not gotten any response as of yet, I got my tester out and found a combination that gives me 12 volts DC, is this what I should put to the mother board ?

Who is the manufacturer of the printer, and what board is it? Easier if we have some more information.

I have a Euroway Prusa i3, a generic knockoff of a Geeetech Pro B. It uses a GT2560 board just for reference info.

the mother board is a “ANET v1.0” and looks like (see pic)
